CJC-1295

CJC-1295 is a long-acting analog of growth hormone-releasing hormone (GHRH). Unlike many research peptides, it has published randomized, placebo-controlled human pharmacology studies demonstrating prolonged stimulation of GH and IGF-1 in healthy adults.

EVIDENCE MAP

What kind of evidence exists?

CJC-1295 has stronger direct human pharmacodynamic evidence than most compounds in this index, though that does not equal approval for consumer use.

Human randomized evidenceDouble-blind ascending-dose trials measured GH, IGF-1, pharmacokinetics, and tolerability.
Biomarker researchProteomic work examined serum changes after CJC-1295-mediated GH/IGF-1 activation.
Mechanistic basisThe compound acts as a long-acting GHRH analog rather than directly replacing growth hormone.
Translation limitsDemonstrating hormone changes in healthy volunteers does not establish benefits for bodybuilding, anti-aging, or other consumer claims.
i

This is a good example of why endpoint discipline matters.

The human trials clearly establish prolonged changes in GH and IGF-1. They do not establish every downstream performance, body-composition, or longevity claim made online.

HUMAN · RANDOMIZED CONTROLLED

Prolonged stimulation of growth hormone (GH) and insulin-like growth factor I secretion by CJC-1295, a long-acting analog of GH-releasing hormone, in healthy adults

2006 · PMID 16352683 · DOI 10.1210/jc.2005-1536

Two randomized, placebo-controlled, double-blind ascending-dose trials found dose-dependent increases in GH lasting at least six days and IGF-1 lasting roughly 9–11 days after a single dose; the estimated half-life was 5.8–8.1 days.

HUMAN · PROTEOMICS

Activation of the GH/IGF-1 axis by CJC-1295, a long-acting GHRH analog, results in serum protein profile changes in normal adult subjects

2009 · PMID 19386527

Serum from 11 healthy young men was analyzed before and one week after CJC-1295 exposure to identify protein-profile changes associated with GH/IGF-1 axis activation.
